چکیده

Summiary. Parameters of the bioassay based on the gibberellin-indluced reducing suigar release of barley endosperm were investigated. Procedures for the rapid handling and processing of utp to several hundred treatments witholut loss in sensitivity of the test are described, and the effects of variations in many aspects of the bioassay were assessed. In general, the variations in varieties, techniquies, additives, conditions, and even gibberellins, all illustrate the stability, sensitivity, and adaptability of the hormoneind(uiced respoiise anid emphasize its uttility as a gibberellini bioassay.

Similarities between gibberellins and related compounds in inducing Acid phosphatase and reducing sugar release from barley endosperm.

Barley endosperm halves release acid phosphatase in response to several gibberellins and gibberellin precursors. Seed halves incubated with 10(-7)m GA(3) at 29 degrees begin to release phosphatase after 11 hr and release it for another 26 hr in response to GA(3). After 37 hr, the rate of release slows to that of seed halves incubated without GA(3). GA(3) is active at 10(-10)m and maximally acti...

Determination of Endogenous Gibberellins in Germinating Barley by Combined Gas Chromatography-Mass Spectrometry

This study was undertaken to elucidate the dominant endogenous gibberellins (GA) and the changes in their levels in germinating barley during malting. Also, a method for determining gibberellins in barley and malt by combined gas chromatography-mass spectrometry (GC-MS) was developed. Endogenous gibberellins in germinating Fuji Nijo I I and Betzes barley were sequentially extracted with ethanol...

Physiological Effects of Gibberellic Acid. X. Gibberellin-Like Substances by Germinating The Release of Barley Embryos

Sum,itmary. Barley embryos, completely free from endospernm, wvere excised from germinating grain at various times and al'lowed to diffuse into an aqueous medium for varying lengths of time. At the end of this time, the embrvos and ambient solutioins were separately extracted. Gibberellin-like activity in the extracts was determined with the barley endosperm bioassay using seed from the same va...

Gibberellic acid-controlled synthesis of alpha-amylase in barley endosperm.

Because the endosperm half of a barley seed produces several hydrolytic enzymes in response to added gibberellins (1, 4, 5, 8), it is an attractive experimental system for a study of the mechanism of action of gibberellic acid. The only living cells in the half seed are those of the aleurone layers. These are an apparently homogeneous collection of respiring, nondividing cells which have the hi...

Aleurones from a Barley with Low [alpha]-Amylase Activity Become Highly Responsive to Gibberellin When Detached from the Starchy Endosperm.

The physiological and molecular bases for contrasting [alpha]-amylase phenotypes were examined in germinating seeds of two barley (Hordeum vulgare L.) cultivars, Morex and Steptoe. Morex is a high-quality malting barley that develops high [alpha]-amylase activity soon after germination. Steptoe is a feed barley that develops only low [alpha]-amylase activity levels during this period. The expre...
